At the end of the 2019, a virus named Severe Acute Respiratory Syndrome Coronavirus (SARS- Cov-2) detected in Wuhan, China. Until November 2021, diagnosis of more than 247 million people and death of 5.01 million people patients due to Cov-2 virus.
Although vaccines and treatments are going on , side by side diagnosis of Covid-19 virus in several methods are under process. There are many serological and nucleic acid amplification test are
done, but some specific test are also there for accurate result.
The various Tests are as follows:
ddRT-PCR-
- This technology is the third-generation RT-PCR method and not requires the standard curve line to achieve absolute quantitative detection of DNA.
- It greatly improves the accuracy of diagnosis, especially for the detection of samples with low copy SARS-CoV by using droplet technology.RT-LAMP–
RT-LAMP-
- LAMP is a new isothermal nucleic acid amplification method with great efficiency.
- The LAMP technology does not need high-priced equipment and reagents with the high advancement of saving time.
- It’s very easy to observe the results for using the Gel Electrophoresis method. So, this is a cost- effective method.
- It has exponential amplification feature and six particular target sequences diagnosed by four separator primers.
- Within one hour and under constant temperature (60–65°C), the templet can be amplified to 10 9 ~10 10 times.
- The results of the LAMP can be easily judged by the turbidity or color of the solution.
CRISPRE–
- The test detection method using enzyme Cas13a/C2c2 based on clustered regularly interspaced short palindromic repeats is a fast, cheap and highly sensitive diagnostics tool.
- The Cas13a detects the target RNA sequence and become active state(Under the guidance CRISPR RNA).When the target RNA sequence is cut, it emits a fluorescent signal.
- This method is easy to operate .It can test quickly without the need for professionals, reducing the shortage of professionals in the large scale fight against COVID-19.
- Beside the methods, other RNA-based method for the detection of SARS-CoV-2 is microarray. It is a rapid and high- throughput method for diagnosis of COVID-19. cDNA is first
produced by coronavirus RNA template. Then the targets are hybridized to the probes. Remove free DNAs by washing the solution and finally probes identify SARS-CoV-2 RNA.
